Inverted Microscopes

Objectives below the stage, and the problems that arrangement solves.

An inverted microscope puts the objectives beneath the stage and the illuminator above it — the opposite of the upright arrangement. It is not a variant for its own sake; it solves a specific problem.

The problem it solves

Cultured cells grow attached to the bottom of a flask or dish, under several millimetres of medium. An upright microscope would have to look down through all that liquid and through the vessel's lid, and its high-power objectives have working distances under a millimetre. It cannot reach.

Inverting the optics puts the objective directly against the thin, flat vessel bottom — exactly where the cells are. The medium is then above the specimen, out of the light path.
